So I know you can format the current date with something like ‘#date.localyyyy-MM-dd H:mm:ss#’ which formats to ‘2017-01-22 17:05:01’.

Is there any way to do this with total game time? Like in a save file or in a Player Menu so I can have "HH:mm" to get a time like 03:45 for 3 hours and 45 minutes played?

Currently not, game time is formatted with hours, minutes and seconds.
